From iPSC discovery to commercial reality: Manufacturing lessons for enabling clinical and commercial success

Two decades following the discovery of induced pluripotent stem cells (iPSCs), the field of stem cell therapies has transformed from a scientific breakthrough into a promising therapeutic platform. Despite the significant advances in the field, the success of iPSC-based therapies ultimately depends on the ability to establish robust, scalable, and commercially viable manufacturing processes. This presentation reflects on some of the key manufacturing advances that have helped move iPSC technologies closer to clinical and commercial reality.
In 2015, we manufactured the first GMP iPSC master cell bank. We then used detailed characterization techniques (e.g. whole genome sequencing) to assess Haplobanks’ genomic stability and safety of iPSC-derived cell therapies. The human iPSCs manufactured under cGMP retained clinical potential, differentiating into all three embryonic germ layers via controlled induction. Importantly, the cGMP iPSCs maintained their long-term stability five years post cryopreservation without acquiring karyotype abnormality, loss of pluripotency, and telomerase activity. We have successfully scaled iPSC expansion and directed differentiation in large scale (3L-50L) bioreactors using computational fluid dynamics (CFD) modeling. This is a major step towards establishment of commercially viable iPSC based therapies. Furthermore, we have highlighted the importance of using automated analytical methods to establish a robust and reliable clinical manufacturing process.
As the iPSC community celebrates twenty years of innovation, the next phase of progress will depend not only on biological discovery but also on the bioprocessing considerations that enable scale, consistency, quality, and commercial viability. This presentation will summarize our key findings around establishing robust GMP banking, phase appropriate automation, characterization and advanced bioprocessing strategies as the foundation for industrialization of iPSC-derived therapies.
